This volume is a record of a meeting entitled "Heparin and Related Polysaccharides" that was held at the Biomedical Center, Uppsala, Sweden between 1-6 September 1991. The meeting was hosted by U. Lindahl, L. Kjellen and I. Bjork, who were helped in their preparations by a scientific advisory panel that included U. Abildgaard, B. Casu, E. Holmer and D. Lane. Altogether, 230 participants from 18 countries attended the meeting, and most were present to be included in a photograph, which is to be found at the end of this volume. The selection of presentations for inclusion in the Symposia of the meeting was made on the basis of the known high quality of the work of the individuals or groups involved. This, we believe, is reflected in the contents of the enclosed articles, which collectively give a comprehensive overview of the present state of knowledge of heparin and related compounds. Some of the areas covered are evolving or controversial and the views expressed in each article should be regarded as those of the author(s). The authors have taken various amounts of care to define all their abbreviations and some familiarity with the different forms of nomenclature used in the fields of polysaccharide chemistry and of the coagulation proteinases and their inhibitors will assist the reader.

This volume focuses on solution and solid-state NMR of carbohydrates, glycoproteins, glyco-technologies, biomass and related topics. It is estimated that at least 80% of all proteins are glycoproteins. Background: There is increased interest in the development of heparin and related polysaccharides for clinical indications beyond their current anticoagulant remit. These indications range from anti-inflammatory and anti-complement agents, anti-metastatic therapeutics, prevention of protracted labour and easing pulmonary conditions such as chronic pulmonary obstructive disease and asthma. Anticoagulant activity may not be necessary and may cause adverse side effects when heparin is used for these conditions. Removal of anticoagulant activity such as selective de-sulfation have been carried out by a few pharmaceutical manufacturers of heparin and some of these products are now in clinical trials. Currently there is limited information on the structure/function relationships between nonanticoagulant heparins and these indications. The aim of this project was to study the structure and function relationship between heparins for some of these new indications. Methods and results: Heparin materials with different structures were prepared and characterised using a range of physicochemical and biological assays. Three different nonanticoagulant functions of these heparin were investigated: 1/ Potential pro-inflammatory properties in the kallikrein-kinin system were investigated for sulfate modified heparin and chondroitin sulfates. Of the samples investigated only oversulfated chondroitin sulfates significantly increased generation of kallikrein and bradykinin, indicating a high degree of sulfation is required to stimulate the kallikrein-kinin system.

Heparins remain amongst the most commonly used drugs in clinical practice. Almost 100 years have passed since the initial discovery of this complex substance and, during this time, understanding of the nature and uses of heparin and related molecules has grown dramatically.
